Tersus polymerase and Tersus Plus PCR kit

cat.# PK123S, PK123L, PK121

- High fidelity
- High specificity
- Fast antibody-based hot start
- TA cloning compatibility

Tersus polymerase is a specially developed mix of proofreading and highly processive PCR enzymes and hot start antibodies that inhibit polymerase activity at room temperature, preventing non-specific amplification and allowing flexible reaction setup. Tersus polymerase has about 4 times lower error rate than Encyclo polymerase that makes it an ideal choice for cloning and other applications requiring high-fidelity amplification. High specificity of Tersus polymerase en-sures its excellent performance in amplification of difficult templates, such as highly homologous repeats, genomic DNA or cDNA libraries.

Tersus polymerase provides robust amplification of DNA fragments up to 3 kb in length.

- Amplification of DNA for subsequent cloning or sequencing
- Site-specific mutagenesis
- High specific PCR from complex templates
- Real-time PCR with intercalating dyes (SYBR Green I etc.)

Tersus polymerase provides highly specific amplification of difficult templates.

A 230 bp fragment of L1Hs repeat was amplified from 5 ng of human genomic DNA using different polymerases.
M - 1 kb DNA ladder; 1 – Taq polymerase; 2 – Encyclo polymerase; 3 – Tersus polymerase, 28 cycles.

PCR amplification of phage λ DNA fragments using Tersus polymerase.

DNA fragments were amplified from 25 ng of template in 15 cycles (wells 1 and 2) and 19 cycles (wells 3-5).
1 – 3 kb fragment; 2 – 5 kb fragment; 3 – 8 kb fragment; 4 – 10 kb fragment; 5 – 15 kb fragment; M – 1 kb DNA ladder.

Possible limitations:

- Not recommended for amplification of long (>3 kb) templates (use Encyclo polymerase instead)
- Not suitable for real-time PCR methods based on DNA probe breakdown (such as TaqMan) due to the lack of 5’>3’ exonuclease activity
- Not recommended for allele-specific SNP detection due to the presence of proofreading 3’>5’ exonuclease activity

Notice to Purchaser:

The Product is intended for research use only. PCR is the subject of patents issued in certain countries. The purchase of this kit does not include a license to perform PCR. However, many researchers may not be required to obtain a license. Other investigators may already have a license to perform PCR through use of a thermal cycler with the appropriate label license.
